ZONK1901-14 — A Serendipitous Discovery to Conquer Inevitable Prostate-Cancer Resistance

Resistance is the inevitable journey of prostate cancer: nearly all patients with metastatic castration-resistant disease ultimately escape current standard-of-care therapy, because single-mechanism agents such as abiraterone (androgen-synthesis inhibition) and enzalutamide (AR antagonism) leave an open route for pathway switching.

ZONK1901-14 is a First-in-Class, dual-active-metabolite prodrug designed from D4A — the Nature-reported multi-target metabolite of abiraterone — that releases two synchronized active warheads in vivo: D4A (a 4-target androgen-pathway inhibitor) and ZONK1901-2, a novel compound more potent than D4A itself. Co-administered to peak together at 4 h with a matched 1.43:1 AUC ratio, they deliver a natural multi-target anti-resistance blockade with no single escape route. In head-to-head xenograft studies at equimolar doses, ZONK1901-14 achieved a 54% tumor growth inhibition — superior to enzalutamide (32%) and abiraterone (30%) — as the only group reaching high statistical significance (P<0.01), while being the only group with body-weight gain (+5.2%), indicating superior tolerability and a wide therapeutic window.

One molecule, two active metabolites, synchronized multi-target blockade — a singular, serendipitous answer to an inevitable resistance.
